pThe mission of the Allen Institute for Immunology is to advance the fundamental understanding of human immunology through the study of immune health and diseases where excessive or impaired immune responses drive pathological processes. The Institute will employ a multi-disciplinary team approach in collaboration with academic centers of human immunology to stronggenerate novel mechanistic insights/strong into the immune synapse in health and in diseases such as autoimmunity or oncology. The Institute will simultaneously provide a strongfoundational data set/strong and tools for future immunological research as well as a novel collaboration portal for the broader scientific community./p
pWe are seeking a summer intern to join the Flow Cytometry department for the summer of 2026. They will contribute to the development of a human antigen-specific T cell Activation-Induced Marker (AIM) assay, bridging flow cytometry and cellular immunology techniques. This project focuses on detecting antigen-specific (Flu/CMV) T cell activation using HLA-typed human PBMC samples and specific peptides. Over the 10-week internship, the intern will learn and apply immune cell culture techniques, gain an introduction to flow cytometry and multiplexed proteomic assays and optimize antigen-specific activation conditions to aid in implementation of this assay. The intern will review literature and existing protocols and work with mentors to define time points for cell resting and cell stimulation steps in the assay. Responsibilities include hands-on setup of PBMC cultures, incubation with Flu, CMV and control peptides, antibody staining and fixation for flow cytometry, and MSD multiplex cytokine workflows. This project provides the intern with robust experiences in single-cell immunology techniques and assay design, with outcomes suitable for inclusion in a T cell AIM assay presentation and poster session. This work will help optimize crucial steps in existing T cell AIM assays under development. This assay model, if successfully developed, could be applied to several ongoing autoimmune and oncology projects./p

liAssay design and protocol optimization: Adjust conditions for cell resting and stimulation to ensure robust, reproducible results/li
